SeV986A

Our group utilizes an replication-defective Sendai virus (SeV) as TB vaccine vector, and constructed SeV85AB and SeV986A vaccine. SeV986A expressed a fusion of acute- and latency-stage Mtb antigens (4 antigens). In murine models, it serves as a potent BCG-booster, significantly reducing bacterial burden in both active and latent infection challenges.

Spore-FP1

Spore-FP1 is a subunit vaccine candidate consisting of inactivated Bacillus subtilis spores coated with the FP1 fusion protein and formulated with Poly(I:C) adjuvant. FP1 fusion protein comprises Ag85B, Acr/HspX and the N-terminal domain of HBHA antigen. It is intended as a mucosal boost vaccine in BCG-immunised hosts.

MtbDsigH

The parental candidate is an isogenic mutant in Mtb CDC1551 gene Rv3223c (sigH). We have now generated multiple Mtb double and triple knock-out (DKO, TKO) strains including: DsigHDsecA2DsodA, DsigHDfbpADsapM, DsigHDleuCDpanCD, DsigHDmetA, DsigHDmce4EDmce4F, DsigHDfadD29, DsigHDhadC, DsigHDRv3683, DsigHDmce1A, DsigHDcobM.

BCG::ESAT6-PE25SS

BCG::ESAT6-PE25SS is a recombinant BCG strain generated on the Danish background. The strain was constructed by fusing the ESAT-6-encoding gene esxA to the general secretion signal for the mycobacterial type VII secretion pathway protein PE25. BCG::ESAT6-PE25SS secretes full-length ESAT-6 via the ESX-5 secretion system, which in contrast to ESX-1 is also present in BCG.

BCG w/MK-2206

Parental BCG efficacy is mainly hampered by its low induction of apoptosis along with high secretion of the immunosuppressive cytokine IL-10. We found that the host transcription factor FOXO3 mediates both, BCG-induced apoptosis and inhibition of IL-10 secretion. The novel vaccination procedure consists of induction of FOXO3 activity, during BCG vaccination, using an anti-cancer drug,…
